New Study Reveals How Genomic Thresholds Shape Meningioma Classification and Recurrence Risk

In a landmark study published in Nature Communications, we mapped chromosomal copy number alterations (CNAs) across more than 1,000 meningiomas, the most common primary brain tumor in adults. The team found that the threshold used to define when a chromosomal arm is "lost" or "gained" dramatically influences how tumors are molecularly graded and how well recurrence can be predicted. By systematically varying these thresholds, the study showed that up to 21% of meningiomas shift between low- and high-grade classifications, with optimal predictive accuracy emerging around a 40% arm-length threshold. We also discovered that CNAs tend to cluster into distinct "small" and "large" genomic events, and that recurrent meningiomas acquire progressively larger alterations over time. Together, these findings highlight the critical importance of standardizing genomic thresholds to ensure robust, reproducible molecular classification, a necessary step toward precision diagnostics, better prognostication, and ultimately more tailored therapies for patients with meningioma.
